Zanubrutinib demonstrates superior tolerability and efficacy in terms of prolonged PFS compared to ibrutinib in relapsed/refractory CLL patients. Notably, this benefit in PFS was maintained in the subgroup of patients with TP53 alterated dissease. Together with the ASCEND trial, these data strongly suggest preferential usage of 2nd-generation BTKI over ibrutinib in CLL, particularly in those with cardiovascular comorbidities.
